反相高效液相色谱法测定野生甘草中甘草次酸含量
Determination of glycyrrhetic acid in glycyrrhiza by RP-HPLC.
【摘要】 采用反相高效液相色谱法测定了野生甘草中甘草次酸的含量。甘草次酸采用超声提取,色谱分离柱为XDB-C8柱(150m×4.6mmi.d.,5μm),以甲醇/水(80/20)溶液为流动相(用磷酸将pH值调节为3.7)。紫外检测波长250nm,柱温为室温。线性范围0.01~0.13mg/mL,线性方程A=50.535+31130.2C,相关系数0.9999,最低检测限0.5mg/L,平均回收率101.1%,相对标准偏差1.10%(n=5)。该方法简单、灵敏、准确,为甘草原药材及其制剂的质量控制提供了一个适用的分析方法。
【Abstract】 The glycyrrhetic acid in glycyrrhiza was determined quantitatively by reversed-phase high performance liquid chromatography. The XDB-C8 column (150mm×4.6 mm i.d., 5μm) was used for separation with methyl alcohol/water (80︰2 0) solution as mobile phase. The detection was at 250nm and the column was at room temperature. The linear range was 0.01~0.13mg/mL. A=50.535+31130.2C and r=0.9999. The detection limit was 0.5 mg/L. The average recovery was 101.1% and the RSD was 1.10% (n=5). The method is simple, sensitive and accurate, and offers a suitable technique for controlling the quality of natural licorice roots and medicines made from them.
