离子交换-脉冲积分安培法分离检测氨基糖苷类抗生素
Separation of several aminoglycoside antibiotics with ion-exchange column following with pulsed integrated amperometric detection
【摘要】 用离子交换柱分离氨基糖苷类抗生素妥布霉素、新霉素B和西梭霉素,用脉冲积分安培电化学检测器进行检测。其流动相不同于一般抗生素的离子对色谱法,它配制简单,仅需使用NaOH-水体系;对两种检测电位波形进行了对比,并选取了表现优良的检测电位。方法对妥布霉素、新霉素B和西梭霉素的检出限分别为8、5、47 ng/mL,在0.1~100μg/mL范围内表现良好线性关系(r=0.9990~0.9996)。该方法应用于市售鲜奶的检测,3种氨基糖苷类抗生素的回收率为85.0%~98.5%。
【Abstract】 A new method was developed for detection of aminoglycoside antibiotics of tobramycin,neomycin and sisomycin by ion-exchange column with pulsed integrated amperometric detection.The method was simple enough with the mobile phase of NaOH/H2O and a good detect waveform based on experiments.The detection limits for tobramycin,neomycin and sisomycin were 8、 5、 47 ng/mL respectively;the linear range was 0.1~100 μg/mL(r=0.9990~0.9995).The method was applied to the detection of milk samples and spiked samples with recoveries of 85.0%~98.5% for the three antibiotics.
