反相高效液相色谱法测定豆制品中两种异黄酮甙元的含量

【摘要】 建立了反相高效液相色谱测定异黄酮甙元的方法。采用Hypersil BDS C18色谱柱(250×4.6 mm i.d.,5μm),流动相为甲醇(A)、0.5%乙酸水溶液(B),流速为1.0mL/min,柱温为35℃,检测波长为260 nm,进样量为10μL。结果表明:大豆黄素的线性范围为0.0450~0.3150μg,加标回收率为99.05%,相对标准偏差(RSD)为1.31%;染料木黄酮的线性范围为0.0448~0.3136μg,加标回收率为98.82%,RSD为0.50%。方法可用于豆豉及其它大豆制品中异黄酮甙元的测定。

【Abstract】 A new method based on RP-HPLC for determination isoflavone aglycones has been developed.established.With Hypersil BDS C18(250×4.6 mm i.d.,5μm),the mobile phase was methanol and(0.5%) acetic acid solution,the flow rate was 1.0 mL/min,column temperature was 35℃,UV-detection was set at 260 nm and the sample size was 10 μL.The results show the linear range of Daidzein is(0.0450)0.3150 μg,the recovery is 99.05%,RSD is 1.31%.The linear range of Genistein is 0.04480.3136 μg,the recovery is 98.82%,RSD is 0.50%.The method has been applied to the determination of isoflavone aglycones in lobster sauce and soy products with satisfactory result.
