化学键合聚乙烯亚胺涂层毛细管柱的快速制备及评价
Fast Preparation and Evaluation of Capillaries Covalently Bonded with Polyethyleneimine
【摘要】 以γ-缩水甘油醚丙基三甲氧基硅烷和聚乙烯亚胺,合成了具有三甲氧基硅烷活性基团的聚乙烯亚胺衍生物,并通过热处理使此高分子聚合物键合于石英毛细管内壁,成功制备了聚乙烯亚胺涂层毛细管柱。所制得的涂层柱能有效抑止电渗流和防止蛋白质在柱上的吸附。在pH7·4的缓冲溶液中,碱性蛋白质的平均分离柱效为1·7×105理论塔板数/米,分析结果呈现出较好的重现性。该制柱方法快速简便,成功率高,涂层柱之间重现性良好。
【Abstract】 A derivative of polyethyleneimine was synthesized with γ-glycidoxypropyltrimethoxysilane and polyethyleneimine. Polyethyleneimine coated capillary was successfully prepared by covalently bonded the polymer onto the inner surface of commercial fused-silica capillary by heating treatment. It had been proved that the electroosmotic flow(EOF) can be suppressed and the adsorption of proteins to capillary can be prevented effectively while evaluating the coated column prepared by above-mentioned method. Meanwhile, the coated capillary was applied in protein separation at pH 7.4, the average separation efficiency of basic proteins reached 10~ 5 plates/m. Good reproducibility of separation was also obtained in further study. This coating method possesses the outstanding character of easy preparation, high successful rate and good reproducibility.
