高效毛细管电泳用于二肽衍生物的手性拆分
Chiral Separation of Derivatized Dipeptides by Capillary Electrophoresis
【摘要】 以新型荧光试剂1,2-苯并-3,4-二氢咔唑-9-乙基氯甲酸酯(BCEOC)作为柱前衍生试剂,基于二元手性选择剂协同作用,在胶束电动色谱模式下考察并优化了二肽类衍生物手性分离的实验条件,实现了15种二肽衍生物的手性拆分,分离度均在2.85以上,其中11种在10.38以上,最高分离度达35.29。
【Abstract】 A simple and rapid method for the separation of derivatized dipeptides using 1,2-benzo-3,4-(dihydrocarbazole)-9-ethyl chloraformate(BCEOC) as derivatization reagent by micellar electrokinetic chromatography(MEKC) was developed.The synthesized process of BCEOC was also described.The detection was performed with diode array at 325 nm.Based on cooperative effect of binary chiral selectors,β-cyclodextrin(β-CD) and sodium deoxycholate(SDC) were used and it was found that the resolution and enantioselectivity were superior to either β-CD or SDC alone.The optimum molar ratio of β-CD and SDC was 1∶1.5 at a total molar concentration of 50 mmol/L.The running buffer was 80 mmol/L tris-(hydroxymethyl) aminomethane-phosphate(tris-H3PO4,pH 9.3),20 mmol/L sodium dodecyl sulphonate(SDS) and 2% acetonitrile,(respectively).In this study,a good baseline separation for 15 pairs of dipeptide derivatives was obtained within 25 min with the resolution greater than 2.85,and 11 pairs of them gave the resolution more than 10.38.The best resolution of 35.29 was achieved.In most cases,the D-enantiomer was eluted prior to L-enantiomer for dipeptide derivatives with only an exception being glycine-serine(Gly-Ser) where L-Gly-Ser eluted earlier than D-Gly-Ser.
