松果体及褪黑素对大鼠胸腺trkB表达的影响
Effect of Pineal Gland and Melatonin on trkB Expression of Rat Thymus
【摘要】 目的研究松果体摘除及补充褪黑素对大鼠胸腺trkB表达的影响。方法选用2月龄清洁级SD大鼠,分为正常组、假手术组、松果体摘除组、松果体摘除+褪黑素低剂量组(腹腔注射7.5 mg.kg-1.d-1)和松果体摘除+褪黑素高剂量组(腹腔注射15 mg.kg-1.d-1),术后4,8周取材。应用RT-PCR法、免疫组织化学染色法观察分析松果体摘除及补充外源性褪黑素后胸腺trkB表达的变化。结果松果体摘除后胸腺trkB mRNA及trkB蛋白表达减弱,补充褪黑素后有所恢复,术后8周时高剂量组胸腺trkB mRNA表达恢复正常,低剂量组胸腺trkB阳性细胞数恢复正常;术后4周低剂量组trkB阳性面积恢复正常。结论松果体及褪黑素对胸腺表达的trkB有影响,松果体摘除导致大鼠胸腺trkB表达减少,而补充外源性褪黑素后能逆转该影响。
【Abstract】 Objective To study the effect of pinealectomy(Px) and melatonin(MLT) supplement on trkB expression in rat thymus.Methods The clean rats of 2 months old were divided into five groups: normal group,sham operation group,pinealectomized group,pinealectomy+(low dose MLT group(intraperitoneal injection 7.5 mg·kg-1·d-1 MLT),pinealectomy+high dose MLT group(intraperitoneal injection 15 mg·kg-1·d-1MLT),and the thymus were taken out at 4th and 8th week after operation.Semiquantitative reverse transcription-polymerase chain reaction(RT-PCR) and immunohistochemistry(IHC) were used to observe and analyze the changes of trkB expression in thymus after Px and MLT supplement.Results trkB(mRNA) and trkB protein expression decreased after Px,and trkB mRNA expression recovered to normal at 8th week in high dose group after MLT supplement;the trkB positive areas in low dose group recovered to normal at 4th week after MLT supplement,but the number of positive cells recovered at 8th week in low dose group after MLT supplement.Conclusion trkB expression in rat thymus were influenced by pineal gland and MLT.Px induced decreasing of trkB expression,and exogenous MLT supplementation can reverse the influence.
