金属离子及脲对中华蜜蜂工蜂碱性磷酸酶的影响
Effects of metal ions and urea on the alkaline phosphatase from Apis cerana cerana Fabricius
【摘要】 通过提纯中华蜜蜂工蜂体内的碱性磷酸酶,研究金属离子及脲对碱性磷酸酶的影响.结果表明:Na+、K+和L i+等正一价金属离子浓度为1.0-10.0 mmol.L-1时,对酶活力没有影响;Mg2+、Ca2+、Ba2+浓度为0-5.5 mmol.L-1时,对酶均有激活作用;N i2+、Mn2+、Co2+、Zn2+浓度为0-200μmol.L-1时,对酶有激活作用,而Cu2+在该浓度下对酶有抑制作用;Hg2+、Ag+及Cd2+浓度为0-150μmol.L-1时对酶有抑制作用;脲浓度低于3 mol.L-1或高于3 mol.L-1时,对碱性磷酸酶有变性失活作用.
【Abstract】 The effects of metal ions and urea on the alkaline phosphatase(AKP) from Apis cerana cerana Fabricius were studied.The results were as follows: Na+,K+ and Li+(1.0-10.[KG8]0 mmol·L-1) had no effect on the enzyme activity;Mg2+,Ca2+,Ba2+(0-5.5 mmol·L-1) could activate the enzyme;Ni2+,Mn2+,Co2+and Zn2+(0-200 μmol·L-1) could activate the enzyme,but in this range of concentration,Cu2+ could inhibit the enzyme;in the range of 0-150 μmol·L-1,Hg2+,Ag+ and Cd2+ could inhibit the AKP;the concentration of the urea(lower than 3 mol·L-1 or higher than 3 mol·L-1) inactivated the enzyme.
