甲醛致小鼠肺DNA蛋白质交联和DNA断裂效应的研究
DNA-protein crosslinks and DNA damages of mice lung induced by formaldehyde
【摘要】 目的研究甲醛致小鼠肺DNA-蛋白质的交联(DNA-prote in crosslinks,DPC)和致DNA的断裂作用。方法以小鼠肺为材料,经体外染毒后,采用KC l-SDS沉淀法和单细胞凝胶电泳法检测液态甲醛染毒后肺部提取细胞中DNA-蛋白质交联物的含量及DNA的断裂效应。结果KC l-SDS沉淀法,低浓度的液态甲醛(5μM,25μM)不能引起DNA-蛋白质的交联(P>0.05),较高浓度(125μM,625μM)可以引起明显的DNA-蛋白质的交联作用(125μM,P<0.05;625μM,P<0.01)。而单细胞凝胶电泳法显示甲醛在低浓度(5μM,25μM)时可以引起DNA链的断裂(P<0.01),在较高浓度(125μM,625μM)时可以引起交联作用(P<0.01)。结论甲醛在较高浓度时可以导致明显的DNA-蛋白质交联作用,而在<25μM时以DNA断裂为主。
【Abstract】 Objective To study the effect of formaldehyde inducing DNA-protein crosslinks(DPC) and DNA damages in mice lung.(Methods)Mice lung was taken as experimental material,KCl-SDS assay and comet assay were used to determine the amount of DNA-protein crosslinks and effect of DNA damages induced by liquid formaldehyde in vitro.(Results) KCl-SDS assay showed that formaldehyde could not cause DPC at lower level(5 μM,25 μM,P>0.05),while at higher level(125 μM,P<0.05;625 μM,P<0.01) it caused DPC obviously.According to comet assay,it caused DNA strand breakage(DSB) at low level(5 μM,25μM,P<0.01) and caused crosslinks at higher level 125 μM,625 μM,P<0.01).Conclusion It was considered that formaldehyde might cause DPC at higher level,but at lower level(25 μM),it mostly caused DSB.
