参附注射液对缺血再灌注心肌炎症反应的影响

【摘要】 目的探讨参附注射液对大鼠缺血再灌注心肌组织内核转录因子-кB(NF-кB)活性、细胞间粘附分子-1(ICAM-1)和白细胞介素-6(IL-6)水平的影响。方法健康SD大鼠36只随机分为三组,空白对照组(S组)、缺血再灌注组(I/R组)、参附注射液组(SF组),每组12只。采用钳闭左冠状动脉前降支制备心肌缺血再灌注模型。电镜观察超微改变,免疫组织化学法检测心肌NF-кB活性和ICAM-1水平,ELISA法检测大鼠心肌IL-6水平。结果1.电镜观察超微改变:I/R组部分肌纤维断裂,肌节模糊,线粒体肿胀,部分线粒体膜破裂,核膜完整高度皱缩,染色质浓缩边聚、呈块状;但SF组上述病变程度明显减轻。2.S组和SF组NF-кB活性、ICAM-1和IL-6水平显著低于I/R组(P<0.01),但SF组NF-кB活性和IL-6水平高于S组(P<0.05)。结论参附注射液能抑制缺血再灌注心肌NF-кB活性和降低ICAM-1和IL-6水平,减轻缺血再灌注心肌炎症反应。

【Abstract】 Objective To study the effect of ShenFu injection on the activty of NF-кB and the level of ICAM-1 and IL-6 following acute myocardial ischemic reperfusion.Methods 36 SD rats were randomized into three groups each consisting of 12 rats: sham group(S group),ischemic reperfusion group(I/R group),ShenFu injection(SF group),.Ischemic Reperfusion models were made by ligated left coronary artery.Immunohistochemistry detected the activty of NF-кB and the level of ICAM-1.ELISA method measured IL-6 level.Results Electron microscope change:There were mitochondria swell,rupture of mitochondrial membrane in I/R group,but less change in SF group.The activity of NF-кB,the level of ICAM-1 and IL-6 in S and SF group were significantly lower than those in I/R group(P<0.01),and the activity of NF-кB,the level of IL-6 of myocardial tissue in SF group were higher than in S group(P<0.05).Conclusion ShenFu injection can inhibit the activity of NF-кB,decrease ICAM-1 and IL-6 level,protect myocardial injury after ischemic reperfusion.
