H9N2亚型禽流感病毒M基因的克隆与序列分析
Cloning and Sequencing of M Gene of A/Chicken/Guangxi/ KMⅢ/99(H9N2)
【摘要】 对分离自中国南方的H9N2亚型禽流感病毒A/Chicken/Guangxi/KMⅢ/99(H9N2)的M基因进行克隆和序列分析。序列分析表明,该M基因全长1 027 bp,包含2个读码框,可分别翻译出M1和M22种蛋白质。M1由252个氨基酸残基组成,M2由97个氨基酸残基组成。与不同毒株比较,结果显示,该M基因与A/Chicken/Guangxi/10/99(H9N2)同源性为100%,当A/Chicken/Guangxi/9/99(H9N2)同源性为99%,与A/Chicken/Guangdong/5/97(H9N2),A/Chicken/Guangdong/11/97(H9N2)等中国分离株同源性达98%。
【Abstract】 According to the reported sequences of Mgene from Avianinfluenza virus(AIV),a pair of pri m-ers was designedfor amplification of Mgene of A/chicken/Guangxi/KMⅢ/ 99(H9N2) by RT-PCR.ThePCR products were clonedinto plasmid pMD18-T.The sequence analysis of theinsert fragmentsinthe re-combinant plasmids pMD-Mindicatedthat the Mgene(1 027 bp) share more than 100 %、99 %、98 %identi-ties compared respectively with AIVstrains that GenBank show.Mgene encodes two proteins : M1proteinand M2protein.The M1proteinis 252 aa long.The M2proteinis 97 aa long.The homology between thisamino acid and other H9N2 M1amino acidin GenBankis above 98 %online.
