大鲵遗传多样性分析

【摘要】 以线粒体DNA控制区为分子标记,对汉水流域大鲵(Andriasdavidianus)的遗传多态性进行了研究。在获得的757bp的碱基序列中,有18个多态性核苷酸变异位点,占全部碱基数的2·38%;基因突变以转换为主,有一处插入或缺失;A、T、C、G四种碱基的平均含量分别为31·8%、33·0%、20·9%、14·3%,且A+T的含量(64·8%)明显高于G+C的含量(35·2%)。28个样本检测到8种单倍型,单倍型间的核苷酸变异在0·13%~1·74%之间,单倍型多样性(Haplotypediversity)指数和核苷酸多样性(Nucleotidediversity)指数分别为h=0·8230±0·0440和π=0·00446±0·00122,显示了大鲵驯养群体的的遗传多样性水平较低。

【Abstract】 A research on polymorphism of giant salamander,was conducted by taking their mtDNA control region as molecular marker.A sequence of 757 bp obtained contained 18 polymorphic nucleotide variable loci,accounting for 2.38% of the total number of bases Transition was the predominating gene mutation.There was only one insertion or deletion.The average contents of the four bases A,T,G and C were 31.8%,33.0% 20.9% and 14.3%,respectively.And A+T contents (64.8%) were significantly higher than those of G+C(35.2%).Eight haplotypes were detected in 28 samples.The nucleotide divergence among the haplotypes was between 0.13 % and 1.74 %.The haplotype diversity index and the nucleotide diversity index were h=0.8230±0.0440 and π=0.00446±0.00122,which suggested that the genetic diversity of the giant salamander population was at a rather low level.
