血管紧张素Ⅱ对大鼠胰岛β细胞形态功能的影响

【摘要】 目的:探讨缓慢输注血管紧张素Ⅱ(AngⅡ)后,大鼠胰岛β细胞功能和形态的变化.方法:给予SD大鼠皮下缓慢输注AngⅡ2wk,成模后再分别给予正常盐(8g/L)和高盐(40g/L)饮食,动态观察血压、血糖、血胰岛素变化,并利用免疫组化的方法了解胰岛β细胞形态的变化.结果:AngⅡ输注后2wk,大鼠血压明显升高;停止AngⅡ输注后4wk,实验组血压继续升高,实验+高盐组血压升高更明显,实验+高盐组β细胞面积明显大于对照组(P<0.05);停止AngⅡ输注后12wk,实验组血压比对照组血压仍高,血胰岛素均明显降低,胰岛β细胞面积明显增大(P<0.05),实验+高盐组变化更明显.结论:缓慢输注AngⅡ可引起血压明显升高,可能影响胰岛β细胞结构,导致β细胞胰岛素分泌功能受损.

【Abstract】 AIM: To study the effects of angiotensin II (AngⅡ) on the function and morphology of islets β-cells. METHODS: The SD rats accepted AngⅡ by hypodermic injection with the osmotic minipump for 2 weeks. Hereafter, they were fed with a normal (8 g/L NaCl) or high-salt (40 g/L NaCl) diet respec- tively. The following indexes, including blood pressure, fasting blood glucose, insulin and the morphology of islets β-cells were observed. RESULTS: The blood pressure of rats was signifi- cantly elevated 2 weeks after accepting AngⅡ via chronic hypodermic injection; at the end of 4 weeks, compared with control group, the blood pressure was increased unceasingly and markedly in the rat with AngⅡ infusions; meanwhile, the areas of islets β-cells in the rats of high-salt diet became larger than those of control group (P<0.05); at the end of the 12 weeks, the blood pressure of rats with AngⅡ infusions was higher than that of control group, in addition, except the fasting blood glucose, the fasting insulin was significantly decreased and the areas of islets β-cells significantly increased (P<0.05), and high-salt loading exacerbated the effects of AngⅡ infusion significantly. CONCLUSION: Chronic AngⅡinfusion increases blood pressure of rats, and effects the morphology of islets β-cells, so its secretory function might be damaged.
