洛沙坦对家兔心肌缺血再灌注损伤的保护作用
Protective effects of losartan on myocardiac ischemia reperfusion injury in rabbits
【摘要】 目的研究血管紧张素Ⅱ(AngⅡ)1型受体拮抗剂洛沙坦对家兔心肌缺血再灌注损伤(myocardialischemiareperfusioninjury,MRI)的保护作用。方法结扎家兔左冠状动脉40min而后再灌注2h建立MRI模型并分别给予2、6、20mg/kg的洛沙坦,于再灌注后5、20、40min、1、2h分别检测心功能、肌酸磷酸肌酶(CK)、乳酸脱氢酶(LDH)、血浆和心肌组织AngⅡ、ATP含量的变化。结果MRI过程中左心室收缩和舒张功能降低,血浆和心肌局部组织中AngⅡ含量增加,心肌酶释放增加,心肌细胞内ATP含量减少,洛沙坦明显保护MRI家兔心功能并降低心肌酶的释放和维持心肌细胞内ATP浓度。结论MRI有可能是与AngⅡ有关,洛沙坦可能通过特异性拮抗AngⅡ1型受体来抵御MRI而发挥心肌保护作用。
【Abstract】 Objective To observe the protective effect of losartan, the antagonist of AngⅡ type1 receptor, on myocardial ischemia reperfusion injury (MRI) in rabbits. Methods The MRI model was inflicted by occluding left anterior descending coronary artery for 40 min followed by 120 min of reperfusion with 2, 6, 20 mg/kg losartan administered to different groups respectively. Cardiac function and CK, LDH, ATP and AngⅡ contents in the plasma and heart tissue were recorded and measured at 5, 20, 40 min and 1, 2 h after reperfusion. Results In MRI rabbits, their systolic and diastolic functions of left ventricle was damaged, myocardiac enzyme and AngⅡ in plasma and cardiac muscle were increased and ATP in myocardiac cells was decreased. Losartan obviously antagonized the above changes during MRI. Conclusion AngⅡ may be involved in the process of MRI, and losartan may protect cardiac muscle by antagonizing AngⅡ type1 receptor.
