坐骨神经损伤后肝细胞生长因子受体c-Met在脊髓和背根节的表达及其意义

【摘要】 目的观察坐骨神经损伤后肝细胞生长因子受体c-Met在脊髓和背根节(dorsal root ganglion,DRG)的表达。方法采用免疫组织化学方法结合图像分析技术检测大鼠坐骨神经结扎后1、3、5、7、14、21和28d c-Met在脊髓和DRG内的表达变化情况。结果术后5d DRG c-Met表达开始增加,7~14d达高峰,之后渐恢复正常;损伤后脊髓前角c-Met免疫阳性产物未见明显变化,但脊髓后角免疫阳性面积5d表达开始增加,7~14d达高峰,之后恢复正常。结论c-Met可能参与了神经损伤后的再生过程。

【Abstract】 Objective To observe the expression changes of hepatocyte growth factor (HGF) receptor c-Met in spinal cord and dorsal root ganglion (DRG) after sciatic nerve injury. Methods The sciatic nerve ligation was made in 42 SD rats. On day 1, 3, 5, 7, 14, 21 and 28 after ligation, the expression of c-Met was detected by immunohistochemical method and image analysis technique. Results c-Met in DRG increased at day 5 after sciatic nerve ligation, peaked at 7-14 d, and gradually returned to normal level. No changes of immuno-positive production in spinal ventral horn after injury were observed, but the immnuno-positive area of spinal dorsal horn increased at 5 d, peaked at 7-14 d, then down to normal level. Conclusion c-Met may be involved in the course of nerve regeneration following peripheral nerve ligation.
