大口黑鲈养殖群体遗传多样性的分析
Genetic diversity analysis of largemouth bass(Micropterus salmoides) cultured in ponds
【摘要】 用RAPD技术对养殖的大口黑鲈M icropterus salm oides群体的遗传多样性进行了分析,即用20个随机引物对30个个体的基因组DNA进行PCR扩增,共扩增出2790条DNA片段,平均每个个体扩增出93条带。在检测到的93个位点中,多态位点数为49个,占52.7%,标记的分子量为0.2~3kb。个体间最大的遗传距离为0.4533,最小的遗传距离为0,其中有27个样品间的遗传距离较小(0.16~0),部分样品之间的遗传距离为0;有3个样品的遗传距离较大,表明大口黑鲈间的相似度较高,遗传多样性较少,但也有少数变异个体。30个个体的平均遗传距离为0.0796±0.1265,与胭脂鱼、黄颡鱼、鳑鮍、鲢、草鱼等其它淡水鱼类相比,它们的遗传多样性水平相仿。
【Abstract】 Genetic diversities of 30 individuals of largemouth bass(Micropterus salmoides) cultured in ponds in Guangdong province were analyzed by the random amplified polymorphic DNA(RAPD) technique using 20-nucleotide-long primers.Twenty primers produced 2 790 well-amplified DNA fragments with reproducible banding patterns and average 93 DNA fragments in one individual.Forty-nine of 93 loci detected were polymorphic,amounting for 52.7%.The DNA fragments were from 0.2 kb to 3 kb in size.The genetic distance between two individuals varied from 0 to 0.4533.The genetic distance between two individuals was from 0 to 0.16 within 27 of 30 individuals and the genetic distance among some individuals was 0.The results indicated that genetic diversities of a majority of individuals were less in cultured population which was contributed to artificial cultured and propagation population than in wild population with similar genetic.But there were 3 individuals whose genetic distance was from 0.14 to 0.45.Compared with other individuals the 3 individuals were rather far off,indicating that there were still some variation individuals in cultured population.The variation was enrichment the genetic diversities of population and benefits selective breeding of largemouth mass.The average genetic distance was 0.0796±0.1265 among the 30 individuals, similar with some freshwater fish,such as Chinese sucker(Myxocyprinus asiaticus),Pseudobagrus fulvidraco,Rhodeus ocellatus,silver carp(Hypophthalmichyhys molitrix),and grass carp(Ctenopharyngon idellus).
