全反式视黄酸对体外培养的人体大隐静脉内膜平滑肌细胞增殖与凋亡的影响

【摘要】 目的研究全反式视黄酸(atRA)对体外培养的人体大隐静脉内膜增厚、平滑肌细胞增生与凋亡的影响。方法30小段(1 cm长)人体大隐静脉(HSV),随机分成HSV组、atRA组和对照组(n=10),atRA组和对照组均体外培养14 d。atRA组:培养液中含atRA100μM;对照组:培养液中含同等量无水酒精(atRA溶剂)。对HSV组及培养后的atRA组、对照组标本进行取材固定,M asson染色,免疫组化测定增殖细胞核抗原(PCNA)及TUNEL法检测平滑肌细胞凋亡,计算机图象分析仪测量血管内膜厚度、计算内膜增生指数及细胞增殖和凋亡百分比。结果体外培养人体大隐静脉能产生明显的内膜增生和平滑肌细胞增殖(P<0.05);atRA组内膜厚度、增生指数及PCNA阳性细胞百分数均明显低于对照组(P<0.05)。而凋亡细胞百分数则明显高于对照组(P<0.05)。结论atRA可以抑制体外培养人体大隐静脉内膜增厚及平滑肌细胞增生,促进平滑肌细胞凋亡。

【Abstract】 Objective To investigate the effects of all-trans-retinoic acid(atRA) on proliferation and apoptosis of smooth muscle cell in human saphenous vein organ culture.Methods Thirty segments(1cm) of human saphenous vein were divided into 3 groups: HSV group,atRA group and control group.The human saphenous vein segments in atRA group and control group were maintained in organ culture for 14 days with 100μM atRA and equivalent concentrations of pure ethanol respectively.All 30 segments human saphenous vein were harvested and the level of proliferating cell nuclear antigen(PCNA) was detected immunohistochemically.The smooth muscle cell proliferation was observed pathologically.Results Cultured saphenous vein segments developed significant neointimal formation and marked smooth muscle cell proliferation(P<0.05).The average neointimal thickness index of proliferation and percentage of PCNA positive cells in the atRA group were significantly lower than those of the control group,and the percentage of apoptosis cells in the atRA group was significantly higher than that of the control group(all P<0.05).Conclusion With the treatment of atRA(100μm),the neointimal formation and smooth muscle cell proliferation can be inhibited and the apoptosis of smooth muscle cell in human saphenous vein organal culture can be induced.
