【Abstract】 It needs dehydrating, infiltrating and embedding before observing when observing plant specimens with paraffin sections. Aiming to solve this time consuming process, cryo-sectioning technique was used to observe stem and root structure of two-year-old Rhodiola sachalinensis A. Bor. The operating process is simple and efficient, and clear plates can be acquired within one day. The key step is to select a proper concentration of glycerol, a suitable temperature of freezing and an appropriate way of section-flatting.更多还原
