家蚕5龄幼虫不同时期中部丝腺细胞蛋白质双向电泳分析

【摘要】 对5龄家蚕不同发育时期的中部丝腺细胞蛋白质变化进行分析研究,有利于发现与丝胶蛋白分泌有关的功能蛋白质。采用蛋白质双向电泳和图像分析技术,发现同一区段的家蚕中部丝腺细胞在5龄第2天、第4天和熟蚕期的蛋白质组成具有显著差异:中部丝腺前部区段5龄第2天表达的特异性蛋白质斑点有8个,第4天有16个,熟蚕期有24个;中部区段5龄第2天表达的特异性蛋白质斑点有9个,第4天有33个,熟蚕期有30个;后部区段5龄第2天表达的特异性蛋白质斑点有10个,第4天有7个,熟蚕期有25个。

【Abstract】 Investigation on the protein varieties in the middle silkgland cells of silkworm,Bombyx(mori),during 5th instar stage could find out the functional proteins concerning with the secretion of the sericin protein.Using two dimensional polyacrylamide gel electrophoresis(2D-PAGE) and the image analysis technology,the results indicated that a significant difference in the protein composition of the same part of middle silkgland cells among 2nd day,4th day and the mature stage during 5th instar.Eight special protein spots were expressed in anterior part of middle silkgland cells in 2nd day,16 in 4th day and 24 in the mature stage. In middle part of middle silkgland cells,9,33,and 34 especial protein spots were detected in 2nd day,in 4th day and in the mature stage,respectively.In posterior part of middle silkgland cells,10,7,and 25 especial protein spots were found out in 2nd day,in 4th day and in the mature stage,respectively.
