大鼠脑损伤后脑细胞凋亡及ICE基因表达的变化
Apoptosis and expression of ICE gene after traumatic brain injury in rats
【摘要】 目的研究创伤性脑损伤(TB I)后脑细胞凋亡的发生规律及白介素1-β转化酶(ICE)基因表达的变化,了解其变化规律及内在联系,揭示TB I后继发性脑损伤的发生机制。方法于TB I模型上,运用组织原位标记凋亡细胞及免疫组化技术,观察TB I后1~14天大鼠大脑伤侧皮层、海马、白质等脑区细胞凋亡的发生及ICE基因表达的变化。结果TB I后,伤侧大脑广泛存在凋亡现象,伤后1天即可出现凋亡细胞,3天达高峰,14天恢复正常;TB I后,ICE基因表达增高,其增高的时相与凋亡规律相似,ICE表达与细胞凋亡发生呈正相关。结论细胞凋亡参与了大鼠TB I后脑细胞的死亡机制,凋亡是脑损伤后细胞死亡的一个重要原因;ICE基因在大鼠TB I后脑组织中表达增加,可能参与了TB I后细胞凋亡发生的正向调节。
【Abstract】 Objective To investigate the regularity of apoptosis and changes of ICE gene expression post-traumatic brain injury(TBI) in rats,to understand the mechanisms of TBI,and to provide the theory of a new therapeutic method.Methods Based on the TBI models,using the technique of TUNEL,ICC,the apoptotic cells and the expression of ICE protein were evaluated,the regularity about apoptosis,the changes of ICE gene expression,and the relationships between them were all studied.Results TUNEL staining results showed that apoptotic cells occurred significantly in brain postTBI.Compared to white matter of cerebrum(WM),there were more apoptotic cells in injured cortex(IC) and sector of hippocampus areas(CA).The number of apoptotic cells increased 1d after TBI,arrived its peak 3d after TBI,and became normal 14d after TBI.ICC results showed the expression of ICE protein increased wildly in brain post-TBI,and arrived its peak 2d post-TBI.Compared to the regularity of apoptosis phenomena and ICE protein expression,they were positive pertinency.Conclusion Apoptosis contribute a lot to the neuronal death post-TBI,the expression of ICE has positive pertinency to the phenomena of apoptosis post TBI.
