黑曲霉D2-26乳糖酶分离纯化研究
Study of isolation and purification of lactase from Aspergillus niger D2-26
【摘要】 为从黑曲霉D2-26发酵液中得到单一的乳糖酶组分,以便研究其酶学性质。试验通过采用过滤、超滤、硫酸铵分级盐析、Sephadex G-25脱盐、DEAE-纤维素-32离子强度梯度层析、Sephadex G-100凝胶过滤、DEAE-纤维素-32 pH梯度层析等方法从发酵液中分离纯化乳糖酶,纯化酶液经PAGE电泳鉴定得到单一蛋白条带。为此建立了一套简单易行的黑曲霉D2-26乳糖酶分离纯化方法。
【Abstract】 In order to get a single lactase component from the Aspergillus niger D2-26 fermentation liquor to study the enzyme properties,this study was done by means of filtration,ultra-filtration,ammonium sulfate fraction,sephadex G-25 gel filtration,DEAE ion-exchange chromatography,sephadex G100 gel filtration to isolate and purify the lactase from the fermentation liquor.The purified enzyme identified by PAGE indicated that a single protein band was obtained.The research established a set of simple and feasible ways in the isolation and purification of lactase from Aspergillus niger D2-26 fermentation liquor.
