灵孢多糖对脂多糖诱导的原代中脑小胶质细胞激活及TNF-a mRNA和iNOS mRNA表达的影响

【摘要】 目的观察灵孢多糖对脂多糖诱导的小胶质细胞激活的影响及对TNF-a mRNA和 iNOS mPCNA表达的影响。方法建立中脑原代小胶质细胞的培养,用灵孢多糖预处理30min,再加入脂多糖处理24 h,通过免疫组化检测OX-42的阳性细胞数及RT-PCR检测TNF-amRNA和iNOS mRNA的表达。结果激活的小胶质细胞体积增大,细胞数量增多,TNF-a mRNA和iNOS mRNA 表达增高,但经较大剂量灵孢多糖(100μg/mL,300μg/mL)预处理后,小胶质细胞的激活被抑制,且总的细胞数量显著减少(P<0.01),TNF-a mRNA和iNOS mRNA表达量也降低(P<0.01)。结论灵孢多糖可以减少TNF-a mRNA和iNOS mRNA的表达,可能是由于灵孢多糖的预处理阻断了由脂多糖诱导的小胶质细胞的激活,提示灵孢多糖可能对中脑多巴胺能神经元具有保护作用。

【Abstract】 Objective To investigate the effect of Ganoderma lucidum polysaccharides (GLPS) on the activity of microglia and the expression of TNF-a mRNA and iNOS mRNA in rat primary glia stimulated with lipopolysaccharides (LPS). Mehtods We cultured the rat primary glia, pretreated with GLPS for 30 mins, then treated with LPS for 24 h, and inspected the positive cell number of OX-42 with immunohistochemistry expression of TNF-a mRNA and iNOS mRNA with RT-PCR. Results The value of stimulated microglia expanded and the number of cell increased.The expression of TNF-a mRNA and iNOS mRNA were increased. After pretreatment with much GLPS(100 μg/mL,300 μg/mL) block the activity of microglia and the total number of microglia also decreased obviously(P<0.01). Consequently the expression of TNF-a mRNA and iNOS mRNA was also significant inhibited (P<0.01). Conclusion GLPS could reduce the expression of TNF-a mRNA and iNOS mRNA. Pretreatment with GLPS possibly block the activity of microglia induced by LPS. It suggests that GLPS may have the protective effect of DA neurons.
