肝脂素诱导KG-1细胞凋亡作用的研究

【摘要】 目的探讨肝脂素是否可诱导白血病细胞系KG-1细胞发生凋亡及其最佳药物作用浓度及作用时间。方法应用肝脂素作用于KG-1细胞,采用锥虫蓝拒染法、MTT细胞活性测定法,确定肝脂素作用于KG-1细胞的最佳药物作用浓度和作用时间,以瑞特染色细胞形态学观察法、流式细胞仪检测肝脂素作用前后KG-1细胞DNA分布,并提取肝脂素作用前后KG-1细胞的DNA进行琼脂糖电泳。结果肝脂素诱导后的KG-1细胞细胞形态学呈现凋亡,DNA电泳出现梯状DNA,流式细胞仪检测出现亚凋亡峰。肝脂素作用于KG-1细胞的最佳药物作用浓度为250mg/ml,最佳药物作用时间为用药后10 ̄22h。结论肝脂素可直接诱导KG-1细胞发生凋亡,其分子机制有待进一步探讨。

【Abstract】 Objective To determine whether heplipin can induce KG-1 cells apoptosis and to study the optimal action time and optimal action concentration of heplipin. Methods To get optimal drug action concentration and optimal drug action time KG-1 cells activity was estimated by trypan blue staining and MTT method after heplipin action. Three methods were adopted in this work: wright staining which can observe morphological change of KG-1 cell; flow cytometry method which can analyse DNA distribution of KG-1 cell; genomic DNA swimming which can observe DNA electrophoregram. Results Heplipin can induce KG-1 cells apoptosis in morphology. DNA ladder appear in DNA electrophoregram. Apoptosis sub-peak appears in flow cytometry detection. The most optimal drug action concentration is 250 mg/ml, the most optimal action time is 10~22 hours. Conclusion Heplipin can induce KG-1 cells apoptosis directly. The mechanisms of it need to be studied in further. <
