头孢药物与牛血清白蛋白的相互作用研究

【摘要】 利用光谱技术研究了头孢拉定(Cefradine)、头孢羟氨苄(Cefadroxil)与牛血清白蛋白(BSA)的相互作用机制.由Lineweave-Burk双倒数图,分别求出了在284,290 nm波长激发下它们与BSA作用的结合常数.BSA内源荧光主要由Trp-214色氨酸做贡献.依据F rster非辐射能量转移机制,得出了头孢拉定与Trp-214的距离r284,激发=2.37 nm,r290,激发=2.40 nm;头孢羟氨苄与Trp-214的距离r284,激发=2.35 nm,r290,激发=2.37 nm;证实药物的稳定性及半衰期与结合常数有关.

【Abstract】 The mechanism of the interaction between bovine-serum albumin(BSA) and Cefradine and between BSA and Cefadroxil is studied by the spectra technology.According to those emission spectrum of BSA excited by 284 and 290()nm,the interaction dissociation constants of BSA and drugs are determined from the double reciprocal Lineweave-Burd plots.Trp-214 residue mainly contributes to fluorescence of BSA.According to the theory of Foster’s non-radiation energy transfer,the binding distances between Trp-214 of BSA and Cefradine are also obtained,r(excited by 284()nm) is 2.37()nm,r(excited by 290()nm) is 2.40()nm;So does Cefadroxil,r(excited by 284()nm) is 2.35()nm,r(excited by 290()nm) is 2.37()nm.The ability of stability and halftime of drugs are related to the dissociation constant.
