前列腺癌患者外周血单个核细胞PSA mRNA表达
Expression of PSA mRNA in mononuclear cells of peripheral blood in patients with prostate cancer
【摘要】 目的:检测前列腺癌患者及对照者外周血单个核细胞前列腺特异性抗原(PSA)mRNA表达,并探讨其临床意义。方法:选择39例前列腺癌患者,12例健康体检者作为对照,取5 mL静脉血,肝素抗凝,Ficoll密度梯度离心分离外周血单个核细胞,RT-PCR检测PSA mRNA表达。结果:①39例前列腺癌患者外周血单个核细胞PSA mRNA表达阳性检出率66.7%(26/39),其中血清PSA>20.0μg.L-1者23例,PSA mRNA表达阳性检出率87.9%(20/23);血清PSA≤20.0μg.L-1者16例,PSA mRNA表达阳性检出率37.5%(6/16)。对照组未见阳性表达。②16例血清PSA≤20.0μg.L-1的前列腺癌患者PSA mRNA阳性表达的Gleason评分为:1~4分0/3,5~7分1/8,8~10分5/5,6例阳性者均为中、低分化癌。结论:外周血单个核细胞PSAmRNA的检测在血清PSA>20.0μg.L-1的前列腺癌患者中具有较高的敏感度,是一种可靠的检测方法。血清PSA≤20.0μg.L-1的前列腺癌患者外周血单个核细胞PSA mRNA的检测可以预示微转移的发生。
【Abstract】 Objective To investigate the expressions of prostatic specific antigen(PSA) mRNA in mononuclear cells of the peripheral blood in prostate cancer patients and controls and discuss its clinical significance.(Methods 39 patients) with prostate cancer and 12 healthy men were selected(normal control group).5 mL anticoagulant blood was collected in every individual.The expressions of PSA mRNA in the mononuclear cells of the peripheral blood were measured by RT-PCR after delamination of the monoclear cells.Results ①26 of 39(66.7%) patients had positive PSA mRNA expression,in which(23 cases) serum PSA>20.0 μg·L-1,positive rate of PSA mRNA expression was 87.9%(20/23),while those serum PSA≤20.0 μg·L-1,positive rate of PSA mRNA expression was 37.5%(6/16).②In the 6 cases whose PSA mRNA was positive and serum PSA ≤20.0 μg·L-1,the distribution of the biopsy Gleason were 1-4 score 0/3,5-7 score 1/8,8-10 score 5/5,pathologic findings revealed middle and low differentiation carcinoma in 6 patients with positive expression of PSA mRNA.(Conclusion The) detection of PSA mRNA has high sensitivity in prostate cancer patients whose serum PSA >(20.0 μg·L-1),which is a reliable method.The detection of PSA mRNA in mononuclear cells of peripheral blood can predict the micrometastasis in prostate cancer patients whose PSA≤(20.0 μg·L-1).
