HPLC测定饮用水中藻类叶绿素含量
Determination of Chlorophylls in Drinking Water by HPLC
【摘要】 研究了高效液相色谱法分析叶绿素以及超声波法提取叶绿素的方法.用超声波破碎藻类20 min达到了叶绿素的完全提取.用V(甲醇)∶V(丙酮)=80∶20的混合流动相,并且加入质量分数为0.1%的甲酸,得到了良好的叶绿素a和叶绿素b液相色谱图,用标准曲线法测定了密云水库不同地点的叶绿素浓度.HPLC作为叶绿素的快速分析方法,可以用于水中叶绿素的定量和定性分析,与荧光分析法相比,具有快速,准确的特点.
【Abstract】 The main goal of this research project was to determine the chlorophylls by HPLC.Chlorophylls were extracted using ultrasonic in acetone in 20 minutes.The determination of chlorophylls was achieved by HPLC using the mixture of methanol and acetone(V(methand)∶V(acetone)=80∶20) as mobile phase in the presence of 0.1% formic acid.The total quantity of chlorophyll a and chlorophyll b in the Miyun Reservoir at different collection place were determined by HPLC using standards curves.HPLC can quickly quantitative performance the analysis and determination of chlorophylls in water.
