产絮凝剂细菌的筛选及其培养条件研究
Study on screening and cultivating conditions of strains producing flocculants
【摘要】 从活性污泥、农药厂土壤中分离筛选到8株产絮凝剂细菌,经过复筛得到一株高效絮凝剂产生菌NT-1。该菌产絮凝剂的适宜碳源为蔗糖,适宜氮源为酵母膏,初始pH值7.0~8.0,装液量45~75mL/150mL,接种量3%~6%,摇床转数为150r/min,32℃培养36~72h;产生具有高效稳定絮凝活性的絮凝剂,对高岭土悬液的絮凝率均在90%以上。
【Abstract】 The present paper intends to introduce the authors’ improved method of optimizing the fermentation conditions of strains to produce flocculants.In the paper,8 stubs of strains for this purpose have been sorted and dissolved from the activated sludge and the soil of pesticide-producing works.Strain NT-1 for producing highly efficient flocculant has been obtained by rescreening.It is a gram positive bacillus with the shape of colony on the beef extract and circular peptone medium.The color of this flocculant stub is milk-white,while its surface feels velvet and ropy but its edge is regular.The fermentation conditions have also been optimized in the following way:the optimum carbon and nitrogen sources of NT-1 strain are sucrose and yeast separately.Under the cultivating conditions(with primary pH 7.0-8.0,the liquid pack of 45-75 mL/150 mL,inoculum size being 3%-6%,at 32 ℃,on a shaking table at 150 r/min),strain NT-1 has to be cultivated for 36-72 h.As a result,the strain can produce a highly efficient flocculant,with flocculation ratio of 90% being against kaolin clay suspension.For the given strain,the cultivating conditions are rather simple with its culture time comparatively shorter.In addition,its production keeps unique flocculent activity and thermal stability,with little need for coagulant aid.All the above characteristics will surely be able to contribute to its broad prospective industrial application.
