烧伤大鼠肾脏p38激酶活化和TNF-α含量变化

【摘要】 目的探讨严重烧伤大鼠休克期肾脏组织p38MAKP活化水平以及血浆和肾组织TNF-α含量水平变化。方法清洁级成年雄性Wistar大鼠88只,随机分为对照组,烫伤组和SB组(烫伤+SB203580组,SB203580为p38MAPK特异性抑制剂),正常对照组(0h)8只;烫伤组和SB两组各48只,后两组又分为1、3、6、12、24h五个时相点,用Westernblotting方法检测肾脏组织p38MAKP的活化水平,放射免疫法检测血浆及肾脏组织TNF-α的含量变化。结果烫伤组伤后1h肾组织p38活化水平明显增多,3h达峰值(与对照组比较,P<0.05),6h后活化量明显减少,血浆和肾脏组织TNF-α含量在伤后6h开始增加,12h达峰值(与对照组比较,P<0.05),24h仍较高、未恢复正常,SB组上述指标要比烫伤组显著减少(P<0.05)。结论大鼠严重烫伤后1h肾组织p38MAPK活化明显增加、3h活化水平达峰值,从伤后6h开始血浆和肾脏组织TNF-α的含量水平增加明显、12h为最高,说明p38的活化在肾脏损伤中起到十分重要的作用。

【Abstract】 Objective To investigate the activated levels of p38 mitogen-activated protein kinase (MAPK) in the renal tissue and the content levels of TNF-α in the renal tissue and serum after severe thermal injury.Methods The eighty eight adult health rats were randomly divided in to three groups:control group,burn group and burn+SB group,control group (0 h) is 8 rats,both of burn and SB groups were 40 rats respectively and they were divided into five scheduled time points respectively (1,3,6,12,24hours postburns);We checked and measured the activated levels of p38MAPK in the renal tissue and content levels of TNF-α in the renal tissue and the serum.Results Both of burn group and SB group,there were much more activated levels of p38MAPK at postburn 1 hour,the highest one was at 3 hours in the renal tissue,comparing with control group (P<0.01-0.05).There was the higher content level of TNF-α at postburn 6 hours and the highest one was at 12 hours in renal tissue!and the serum,comparing with control group (P<0.01-0.05).The actived level of p38MAPK and the content level of TNF-α in renal tissue and in the serum of SB group were less than that of burn group (P<0.05).Conclusion There were over activation of p38MAPK at 1 h and 3 h in the renal tissue in rats in the early stage after severe thermal injury,SB could inhibit the activation of p38MAPK and reduce levels of TNF-α in renal tissue and the serum.
