颉颃放线菌XA-1菌株发酵条件研究

【摘要】 对颉颃放线菌XA-1菌株的发酵条件进行初步研究,确定了其最佳发酵培养基组成及培养条件。当马铃薯淀粉为碳源、黄豆饼粉和蛋白胨混合为氮源时,发酵滤液的抑菌作用最强。选用优化的培养基组成,菌株XA-1的最佳发酵条件为26℃,120 r/min,培养基初始pH值为4.5,摇瓶装液量90 ml/250 ml,最佳发酵时间为88 h,培养96 h的种子液以10%的接种量转接有利于提高抑菌活性。经发酵培养基和发酵条件的优化,发酵液抑菌圈达到了34 mm以上,比原始发酵液抑菌圈增大了10 mm,获得了较多的抗生素产量。

【Abstract】 The fermentation conditions of XA-1 strain of antagonistic actinomycetes were studied.The experimental results indicated that the composition of fermentation medium and cultural conditions could improve the inhibition activities of ferment filtrate.The influence of carbon sources and nitrogen sources on the antibiotic of XA-1 strain was different from the single factor test.Potato’s starch was the best carbon sources,and soybean pie flour and peptone were the nitrogen sources.The optimal fermentation condition was fermentable temperature 26 ℃,120 r/min of rotation,initial pH 4.5,90 ml liquid medium in 250 ml flask,seed age 96 h,inoculation volume 10% and culture time 88h. The inhibition diameter of the optimal fermentation liquid exceed 34 mm,which improved 10mm more than that came from original fermentation conditions.
