【摘要】 用MTT法、克隆分析、流式细胞仪及免疫组织化学法 ,观察了小檗碱对人胃癌细胞株BGC 82 3增殖的影响 ,结果发现小檗碱对人胃癌细胞株BGC 82 3有抑制增殖作用 ,且与药物浓度呈正相关。其机理主要为使突变型 p5 3和 p2 1蛋白的表达下调 ,阻滞胃癌细胞进入细胞周期 ,使人胃癌细胞BGC 82 3的细胞周期阻滞于G0 /G1期。更多还原

【Abstract】 To investigate the effects of berberine on proliferation of human gastric carcinoma BGC 823 cell, the effects of proliferation inhibiting with MTT assay, clone inhibiting experiment, flow cytometer and immunohistochemical methods were used. The results showed that berberine could inhibite proliferation of BGC 823 cell and function gets stronger with its concentration increasing, which demonstrates berberine is of anticancer effect in vitro. The mechanism perhaps is downregulation mutant p53 and p21 protein expression, and block human gastric carcinoma cell entrance cell cycle, that block human gastric carcinoma BGC 823 cell cycle in G 0/G 1 phase.更多还原
