异紫堇啡碱抑制血管平滑肌由受体中介的细胞内游离钙的增高
Isocorydine inhibited the receptor-mediated increasing of the cytosolic free Ca2+ level in the blood vessel smooth muscle cells
【摘要】 目的 观察扩血管药异紫堇啡碱 (ISOC)对去甲肾上腺素 (NA)和高钾所致血管平滑肌细胞内游离钙浓度 ([Ca2 + ]i)增高的影响 ,分析该药扩血管作用的原理。方法 利用钙荧光指示剂Fura 2 /AM负载的家兔血管平滑肌培养细胞 ,动态地观察在ISOC存在下 ,由NA和高钾所增加的 [Ca2 + ]i的改变 ,分析ISOC对NA和高钾所致钙内流和 /或钙释放的影响。结果 ISOC对血管平滑肌静息 [Ca2 + ]i无影响 ,该药在 10 -5和 5× 10 -5mol/L时对高钾所致 [Ca2 + ]i的增高也无明显抑制 ,但能明显抑制NA所致细胞 [Ca2 + ]i的增高。结论 ISOC抑制由受体中介的血管平滑肌 [Ca2 + ]i的增高。
【Abstract】 Objective To analyse the vasodilative mechanism(s) of isocorydine(ISOC), its effects on the increasing of the cytosolic free Ca 2+ level ([Ca 2+ ]i) induced by noradrenaline (NA) and high potassium in the blood vessel smooth muscle cells were investigated.Methods The changes of the [Ca 2+ ]i increased by NA and high potassium in the presence of ISOC were determined, using fura-2/AM-loaded cells of the blood vessel smooth muscle of the rabbit.Results ISOC at 10μ mol.L -1 and 50 μmol.L -1 had no effect on the basic [Ca 2+ ]i and the [Ca 2+ ]i increased by high potassium, but inhibited the [Ca 2+ ]i increased by NA significantly.Conclusions ISOC inhibited the receptor-mediated increasing of [Ca 2+ ]i in the blood vessel smooth muscle cells.
