RP-HPLC法测定人血浆中多索茶碱浓度
Determination of Doxofylline Concentration in Plasma by RP-HPLC
【摘要】 目的:建立RP-HPLC法测定人血浆中多索茶碱浓度,探讨3名支气管哮喘患者口服多索茶碱片的药动学特性。方法:流动相采用甲醇-水-三乙胺-冰醋酸(35:65:0.01:0.01),流速为1md·min-1,检测波长为272nm。检测限为20ng·ml-1。数据用“3p97”程序计算机处理。结果:多索茶碱低、中、高3个浓度(0.04μg·ml-1,0.66μg·ml-1及2.65μg·ml-1)的回收率分别为(95.85±3.69)%,RSD=3.9%,(101.30± 3.74)%,RSD=3.7%及(100.70±5.84)%,RSD=5.8%;日内精密度分别为0.0403±0.0023,RSD=5.8%,0.6684±0.0247,RSD=3.7%及2.5426±0.0979,RSD=3.9%;日间精密度分别为:0.0428±0.0028,RSD=6.5%,0.6673±0.0265,RSD=4.0%及2.5554±0.0868,RSD=3.4%。3名患者有1名是两室模型,2名为一室模型。结论:本法简单、快速、分离度好,最低检测限能满足临床要求,所用试剂廉价易得,可用于治疗药物监测。
【Abstract】 Objectives: To establish a method to determine doxofylline concentration in plasma by HPLC, and its pharmacokinetic properties were studied. Method; The mobile phase was composed of methol-water-triethylamine-acetic acid glacial (35:65:0.01: 0.01), flow rate was 1 ml min-1, the detecting wavelength was272 nm and the limit was20 ng ml-1. All data were dealt with by "3p97" program. Result; The recoveries of three doxofylline concentrations were 95. 85 ± 3.69% with RSD = 3. 85% , 101.30 = 3.74% , RSD = 3.69% , and 100.70 ± 5.84% with RSD = 5.80% , respectively. In the three patients, one was suitable for two-compartments model, the others were suitable for one-compartment model. Conclusion; The method is found to be rapid, accurate and precise. It is suitable for clinical pharmacokinetic study.
