【Abstract】 Objective The purpose of this study was to investigate the role of NO (nitric oxide)in experimental hepatic fibrosis.Methods Experimental hepatic fibrosis in rats was induced by intraperitioneal injecting of thioacetamide(TTA). Rats were divided into control group、TAA group and TAA+N~G-nitrol-L-arginine methyl ester (L-NAME)group. TAA group and TAA+L-NAME group rats were injected TAA(100mg/kg ip)and TAA+L-NAME (200mg/kg ip). After 13 weeks, plasma endotoxin content、serum alanine transaminase(ALT) serum LDH activity and plasma nitric oxide (NO) and hepatic tissue NO、collagen were measured. The changes of hepatic histology were observed by HE and V· G stain.Results Liver injury and liver fibrosis were generated in rats with TAA(ip). The results indicated that ALT、LDH、LPS、collagen and NO contents in plasma and hepatic tissue NO contents were increased significantly in TAA group (vs control group P<0.05). Those indices were higher in TAA+L-NAME group than those in TAA group. HE and V·G stain showed that the grade of hepatic fibrosis and hepatic injury were more serious in TAA+L-NAME group than those in TAA group.Conclusion Nitric oxide plays a protective role in the process of hepatic fibrosis.
