川芎嗪和甘利欣对瘢痕成纤维细胞增殖和Ⅰ、Ⅲ型前胶原基因表达的影响

【摘要】 目的观察川芎嗪、甘利欣对体外培养瘢痕成纤维细胞增殖和Ⅰ型、Ⅲ型前胶原基因表达的影响,探讨它们治疗瘢痕疙瘩的机理。方法体外培养瘢痕成纤维细胞。应用不同浓度的川芎嗪、甘利欣作用于3~9代瘢痕成纤维细胞。MTT法测定川芎嗪、甘利欣对瘢痕成纤维细胞增殖的影响。核酸斑点杂交法检测川芎嗪、甘利欣对瘢痕成纤维细胞Ⅰ型、Ⅲ型前胶原基因表达的影响。结果MTT法显示,川芎嗪(0.1mg/mL,0.25mg/mL)、甘利欣(0.1mg/mL,0.25mg/mL,0.5mg/mL),作用时间48h、72h,均能明显地抑制瘢痕成纤维细胞增殖。随浓度增加,川芎嗪(0.5mg/mL,1.0mg/mL)、甘利欣(1.0mg/mL),在24h、48h、72h均显示明显的抑制作用(P<0.05)。浓度为1.0mg/mL、作用48h,72h,两种药物的抑制作用与曲安奈德组相比无显著性差异(P>0.05)。核酸斑点杂交结果提示,川芎嗪、甘利欣可显著降低瘢痕成纤维细胞内Ⅰ型、Ⅲ型前胶原mRNA的含量(P<0.01)。结论川芎嗪、甘利欣对体外培养的瘢痕成纤维细胞的增殖有显著的抑制作用,呈明显的剂量依赖关系和时间依赖关系。川芎嗪、甘利欣显著抑制瘢痕成纤维细胞内Ⅰ型、Ⅲ型前胶原mRNA的表达。

【Abstract】 Objective To investigate the effects of Tetramethylpyrazine(TMP )and Diammonium Glycyrrhizinate(DG)on keloid fibroblasts(KFB)proliferation and the mechanisms of inhibitory effects of TMP and DG on collagen sythesis in keloid-derived fibroblasts.Methods Keloid fibroblasts were isolated from fresh keloid tissue and cultured.The KFB(from the3rd to9th passages)were treated with various concentrations of TMP and DG.Triamcinolone acetonide was used as a positive control,cell culture solution as negative control.The effects of TMP and DG on KFB proliferation was examined by MTT colorimetric cell proliferation assay.Dot blot hybridization was used to detect the expressions of typeⅠandⅢprocollagen mRNA in KFB.Results Both TMP and DG at lower dose(0.1mg /mL,0.25mg /mL,0.1mg /mL,0.25mg /mL and0.5mg /mL respectively)after treating48h and72h had significant inhibition effects(P<0.05,P<0.01).At higher dose(TMP 0.5mg /mL&1.0mg /mL,DG1.0mg /mL),their signicant inhibition effects on KFB proliferation was observed at any detected time .The inhibition effects of both TMP and DG on KFB proliferation is similar to that of triamcinolone acetonide(P>0.05)at1.0mg /mL after48h,72h.Dot blot hybridization used indicated that both TMP and DG could significantly ruduce the centents of th eⅠandⅢprocollagen mRNA in the KFB(P<0.01).Con clu-sion TMP and DG can inhibit KFB proliferation and collagen synthesis at transcriptional level in vitro.
