辛伐他汀对骨髓基质细胞骨形成蛋白-2表达及碱性磷酸酶活性的影响

【摘要】 目的 研究辛伐他汀对原代培养的骨髓基质细胞骨形成蛋白 - 2 (BMP- 2 )表达及碱性磷酸酶活性的影响 ,探讨其刺激成骨的作用机制。 方法 体外培养成年小鼠骨髓基质细胞 ,以不同浓度 (0、0 .1、 0 .2、0 .5和 1 .0μmol/L )的辛伐他汀、基因重组人 BMP- 2 (1 0 0 ng/ml)作用 72小时碱性磷酸酶活性的酶学测定 ;免疫细胞化学染色 ,Western Blotting检测 BMP- 2表达的变化。 结果 辛伐他汀作用 72小时后 ,细胞中的 BMP- 2表达水平增高 ,随浓度增加 ,表达量增加 ,对照组少量表达 ;碱性磷酸酶活性显著增加 ,呈剂量依赖关系 ,辛伐他汀 0 .5和 1 .0 μmol/L与对照组间具有统计学意义 (P<0 .0 5)。 结论 辛伐他汀可促进骨髓基质细胞中 BMP- 2的高表达 ,细胞自分泌或旁分泌BMP- 2增多 ,碱性磷酸酶活性增高 ,有促进成骨的作用

【Abstract】 Objective To study the effect of simvastatin on the expression of bone morphogenetic protein 2 (BMP 2) and alkaline phosphates (ALP) activity in the primary cultured bone marrow stromal cells, and to elucidate the mechanism of the anabolic osteogenetic effect of simvastatin. Methods Bone marrow stromal cells in femur and tibia of adult mouse were cultured in vitro. After treated with different concentrations of simvastatin (0, 0.1, 0.2, 0.5 and 1.0 μmol/L) or recombinant human BMP 2 for 72 hours, ALP activity of bone marrow stromal cells was determined. BMP 2 expression of bone marrow stromal cells was analyzed by using immunocytochemistry and Western blotting. Results After treated with simvastatin for 72 hours, BMP 2 expression increased, while little BMP 2 expression could be observed in the control group. ALP activity also increased in a dose dependent manner; t test showed that ALP activity in the group which concentrations of simvastatin were 0.5 μmol/L(t=2.35, P =0.041), 1.0 μmol/L(t=2.348, P =0.041) had significant difference when compared with control group.Conclusion Simvastatin lead to high expression of BMP 2 in bone morrow stromal cells, via the increased auto or para crine of BMP 2, and ALP activity increased. These may be parts of the mechanism on the anabolic osteogenetic effect of simvastatin.
