【摘要】 通过 PCR方法克隆了从我国哈尔滨分离的一株鸡贫血病毒 (CAV)的 VP2基因 ,并对之进行了测序 ,该基因的开放读码框由 6 5 1bp组成 ,编码 2 16个氨基酸。通过将本次克隆的基因与 Gen Bank收录的 CAV的 VP2基因比较 ,同源性至少为99% ,未发现与本次克隆的 VP2蛋白完全一致的 CAV分离株 ,与之同源性最好的是 CIA - 1的 VP2蛋白 ,相差 1个氨基酸 ,与Cux- 1也仅相差 2个氨基酸 ,因此从该蛋白看 CAV哈尔滨分离株的变异程度不很高。比较这 2 7株病毒的 VP2及其基因序列 ,发现共有 31处核苷酸发生变异 ,这些变异将导致 VP2的 12个氨基酸变化 ,尽管他们散布于整个 VP2 ,但在 186到 191号氨基酸区域存在一个没有报道过的变异程度相对较高的区域 ,CAV的 VP2是相对较保守的蛋白更多还原

【Abstract】 VP2 gene was amplified by polymerase chain reaction from liver of chicken infected by chicken anemia virus isolated in Harbin of China and cloned into pUC18.The sequence was analyzed with F and R primer.It was found that it was of well homology to 26 VP2 genes in Genbank by comparing Genbank through BLAST.Amino acid of VP2 was analyzed with alignment of the 27 CAVs.It was indicated that there are 12 amino acid differences among 216 amino acid residues of VP2.It was also found that there is a hypervariable region from position 186 to 191 in VP2.It seems that VP2 of CAV is not a optionally mutable protein.更多还原
