【摘要】 目的　环磷酰胺 CTX,甲氨喋呤 MTX,甲基强的松龙 M- pred,秋水仙碱 COLC,刺五加 APS,环孢霉素 A Cs A,观察 6种作用机制不同的免疫抑制剂 ,对人胚肺二倍体纤维母细胞 (2 BS)的影响 ,初步筛选出治疗肺纤维化更有效的药物。方法　采用生物活性法观察 6种不同浓度的药物在不同时间对 2 BS细胞的增殖抑制作用 ,利用流式细胞仪检测药物诱导 2 BS细胞凋亡小体形成百分率及其细胞周期的影响。结果　 1 6种药物对 2 BS细胞均有增殖抑制作用 ,与无药空白对照组比较均有显著差异 (P<0 .0 1 ) ,Cs A作用明显大于其它 5种药物 (P<0 .0 5)。 2 6种药物均有诱导2 BS细胞凋亡作用 ,各种药物组 2 BS细胞凋亡小体形成百分率与无药空白对照组比较差异显著 (P<0 .0 1 ) ,Cs A的作用最强 ,明显高于其它药物。 36种药物对 2 BS细胞的作用呈现一定的时效和量效关系 ,即细胞增殖抑制率 (CIR% )随着药物浓度的增加和时间的延长而增高 ,Cs A作用最明显。4Cs A对细胞整个增殖期均有作用 ,M- pred和 APS作用于 S期 ,COLC作用于 G2 /M期 ,MTX,CTX作用于 G0 /G1期向 S期过渡阶段 ,使 G0 /G1期细胞发生阻滞。结论　实验中的 6种药物均有抑制 2 BS细胞增殖和诱导其凋亡作用 ,其中 Cs A作用最强 ,可能 Cs A为治疗肺纤维化疗效更好的药物?更多还原

【Abstract】 Objectives To select the effective drug for intersticial pulmonary fibrosis in six partial immunodepressives. (CTX, MTX, M-pred COLC, APS and A CsA .Methods The in vitro inhibitive effect of six group drugs at different concentrations on the growth of 2BS cells was evaluated at various time by bioassay. The influence on induction of the apoptosis body ratio was analysed by follow cytometry.Results The proliferative inhibition function and inducing apoptosis of the six kinds of drugs, which went up with the increase of drug concentrations and time , were significant （ P <0.01） and especially CsA （ P <0.05）. MTX, and CTX, noted on the course of G₀/G₁ phase going to sphase and blocked G₀/G₁phase; COLC acted on G₂/M phase, CsA acted on each proliferation phase; M-pred and APS acted on S phase .Conclusions The expectation to treat pulmonary fibrosis was provided with the proliferative inhibition function and inducing apoptosis of the six kinds of immunodepressive especially CsA.更多还原
