VEGF在机械性TMR中促心肌血管再生与重建的实验研究

【摘要】 目的:从形态学角度观察血管内皮细胞生长因子(VEG)在机械性TMR(transmyocardialrevascularization)管道内促血管再生与重建的作用。方法:利用高速空心钻头在家兔左室前壁进行TMR,形成透室壁性心肌管道。实验组的TMR管道内注射VEGF;对照组仅做单纯机械性TMR。动物术后不同时间处死,利用组织切片染色法,对照观察实验组和对照组TMR管道内血管再生与重建的状况。结果:术后2周,实验组和对照组的TMR管道内新生小血管的密度/数量分别为(24.20±0.98)条/10 ×4倍和(8.41±0.59)条/10×4倍,P<0.001;术后6~8周,分别为(11.08±0.96)条/10×4倍和(3.31±0.57)条/10×4倍,P<0.001。结论:VEGF对机械性TMR管道内血管的再生与重建具有非常明显的促进作用。

【Abstract】 Objective: To research the effect of vascular endothelial growth factor (VEGF) on the angiogenesis and revascularization in the drilling TMR channels from the view of morphology. Methods: Transmyocardial channels were made by machine drilling in vivo on left ventricular wall of the rabbits. The control group was treated only with drilling TMR, but the experimental group was injected with VEGF in channels after drilling TMR. The animals in two groups were sacrificed at intervals of 1,2,4 and 6-8 weeks after TMR. The effect of angiogenesis and revascularization were observed, and the new blood vessels in channels of two groups were measured under microscope. Results: Statistical analysis demonstrated a high significant difference between the experimental and control groups in the density and quantity of new vessels (after 2 weeks, 24.20± 0.98 NV per 10 × 4 field in experimental group versus 8.41 ± 0.59 NV per 10 × 4 field in control group, P < 0.001; after 6-8 weeks, 11.08 ± 0.96 NV per 10 × 4 field in experimental group versus 3.31±0.57 NV per 10×4 field in control group, P< 0.001). Conclusion: The results confirm that the good effect of angiogenesis and revascularization in the drilling channels can be received by the stimulation of VEGF.
