硫酸二乙酯和紫外线对金霉素链霉菌的诱变育种
The effect of diethyl sulfate and ultra violet light as mutagenic agents on culture selection of Streptomyces aureofaciens
【摘要】 考察了硫酸二乙酯 (DES)和紫外线 (UV)对金霉素链霉菌进行诱变的效果。其中 2 %DES诱变剂对金霉素链霉菌无明显的致死效应 ,但在含有金霉素 2 2 0 μg/ml的培养基上筛选结果表明 ,DES对金霉素产生菌产生了较强的诱变效果 ,菌株正突变率达 5 2 %。对金霉素链霉菌进行紫外诱变的结果表明 ,在营养贫瘠培养基上筛选的诱变效果较营养丰富培养基上的效果好 ,且遗传特性稳定。
【Abstract】 Two percent of diethyl sulfate (DES) could not produce significant lethal effect on S.aureofaciens , but in culture medium containing chlortetracycline (220μg/ml), an apparent mutation effect was observed , and the positive mutation rate reached 52%. In the case of using ultra violet light as mutagenic agent in the selection of S.aureofaciens , the result was better in poor nutrient medium than in the rich one, and the cultrue hereditary characters were also more stable.
