依降钙素对破骨细胞影响的形态学观察

【摘要】 目的 :观察依降钙素的生物学活性及破骨细胞在其影响下的形态学变化。方法 :用含不同浓度依降钙素的培养液培养Wister乳鼠的破骨细胞及离体胫骨 ,扫描及透射电镜分别观察破骨细胞表面绒毛的变化 ,图像分析仪分析图片。结果 :破骨细胞与骨片的结合力减弱 ,表面绒毛减少。图像分析统计 ,2 6.8× 10 -11Mol/L浓度为最低有效浓度。结论 :一定浓度的依降钙素对破骨细胞形态变化有明显影响

【Abstract】 Objective:In order to understand the reducing mechanism of serum calcium induced by a new artificial elcatonin(derivative of natural calcitonin),we studied the morphological changes of osteoclasts in vitro.Methods:Osteoclasts were isolated from Wistar rat(1 5 days)and incubated on bone sheet in medium RPMI 1640.Elcatonin were added to the medium to different concentration.The microvillus of osteoclasts were observed with scan electron microscopy at different time.The tibia of Wistar rats(1 5 days)were incubated in elcatonin medium and sliced.The microvillus was observed under transmission electron microscopy.Calcitonin produced by Sigma Co.serve as a standard control.Result:In elcatonin medium,the numbers of microvillus on osteoclasts reduced and length of them shortened.And the cells partly separately from bone sheet.Image analysis state that the elcatonin have obvious effect on osteoclasts at the concentration more than 26.8×10 -11 Mol/L.Conclusion:Elcatonin can restrain the bone resorption of osteoclasts by the numbers reducing and the length shortening of microvillus.The effects were similar to standard calcitonin.
