【摘要】 目的 :探讨顺铂对耳蜗毒性的影响。方法 :将豚鼠 12只随机分成顺铂组、对照组 ,每组 6只。顺铂组连续 5d腹腔注射顺铂 ,2ml·kg- 1·d- 1;对照组连续 5d腹腔注射生理盐水 ,2ml·kg- 1·d- 1。两组豚鼠均于实验结束后测耳蜗血流、耳蜗电图 ,断头 ,取耳蜗 ,用不同方法染色固定 ,铺片及电镜观察。结果 :顺铂组动物在给药后体重减轻 ,耳廓反射减退 ,耳蜗血流下降 ,阈值提高 ,毛细胞大片缺失 ,两组比较 ,差异有显著性 (P <0 .0 1)。顺铂组血管纹较对照组血管纹明显扩张 ,红细胞聚集。扫描电镜下观察 ,顺铂组的绝大部分内、外毛细胞的纤毛散乱 ,倒伏 ,甚至缺失 ;透射电镜下观察 ,顺铂组血管纹线粒体水肿变性。结论 :顺铂对耳蜗有损坏作用。更多还原

【Abstract】 Objectives:To study the effect of cis-platin on cochleas.Methods:The twelve guinea pigs were divided randomly into two groups: a cis-platin group, a control group. The control group was injected to abdominal cavity with physiological saline (2ml·kg -1 ·d -1 ) for successive five days. The cis-platin group was injected to abdominal cavity with cis-platin (2ml·kg -1 ·d -1 ) for successive five days. All the guinea pigs were killed at the time just after the experiment was completed and cochlear blood flow and CAP were tested. The cochleas were removed and fixed in different fixatives. The specimens were sent for surface preparing, outer hair cell counting, electron microscopy scanning and electron microscopy transmitting respectively after different disposals.Results: The cochlear blood flow was lowed,auditory sheld was bigger ,injury of hair cells was serious on cis-platin group ,while that of a control group was normal . There was significant difference between two groups (P<0.01). Stria vascularis enlarged and red cell gathered in the cis-platin group. Under scanning electron microscope, the ciliums of the majority inner hair cells and outer hair cells in the cis-platin group lodged, even disappeared, Under transmission electron microscope, about stria vascularis in cis-platin group, the edema and denaturation of mitochondria was very obvious. Conclusions:We can draw conclusion that cis-platin affects cochlear.更多还原
