弓形虫病原基因检测用于孕妇感染及感染后对胎儿危险性的研究
STUDIES ON THE DETECTION OF TOXOPLASMA INFECTION IN PREGNANT WOMEN BY PCR/DNA PROBE AND THE HARMFULNESS OF THE INFECTION ON FETUS
【摘要】 目的 研究弓形虫病原基因检测方法及孕妇感染弓形虫后对胎儿的危害作用。 方法 根据弓形虫基因序列 ,设计合成 PCR扩增引物 ,研究 PCR/DNA生物素探针检测试剂 ,并应用该试剂检测异常妊娠孕妇的血液和流产物 ,同时设立对照检查。 结果研制的弓形虫检测试剂具有良好的敏感性和特异性 ;在异常妊娠收集的病态流产物的检测中 ,阳性率为 2 9.3% ,分类比较中以死产死胎的阳性检出率最高 ,为 36 .3%。血样的检测阳性率为 2 7.0 % ,而正常对照流产物和血样的检测阳性率分别为 3.2 %和 0。 结论 研制的试剂及检测方法可用于孕妇虫血症的病原检测 ;孕妇弓形虫感染对孕期胎儿具有直接的危害作用。
【Abstract】 Objective To study on the detection of Toxoplasma infection in pregnant women using gene method and to evaluate the harmfulness to fetus after mother infection. Methods According to gene sequencing, the kit of PCR/probe biotin to detect Toxoplasma has been prepared and been used to detect samples of blood, morbid abortion and controls. Results The kit was very specific and sensitive in this experiment. The detectable DNA volume of the method is 5 parasites/μl. There was no cross reaction with other non Toxoplasma DNA. Of 99 morbid abortion samples detected, 29 was positive with positive rate of 29.3%. The highest positive rate was samples from stillborn fetus with positive rate of 36.3%. 170 blood samples from women after morbid abortion were detected, and the positive rate is 27%, The control blood samples and non morbid abortion samples were also detected with the positive rates being 0 and 3.2% respectively. Comparison results of the PCR/gel electrophoresis and the PCR/probe showed that the latter was good not only in sensitive, but also in repetitive. Conclusion The kit is useful for detection of Toxoplasma infection. The Toxoplasma infected in pregnant women could harm to fetus directly.
