阻断共刺激分子B7-1和B7-2对感染蠕虫小鼠免疫应答的影响
THE EFFECT OF INHIBITING THE COSTIMULATORY MOLECULES B7-1 AND B7-2 ON THE MICE IMMUNE RESPONSE TO HELMINTH INFECTION
【摘要】 目的 探讨共刺激分子 B7- 1和 B7- 2在蠕虫感染免疫中的作用。 方法 用 BAL B/ c小鼠皮下注射感染巴西日圆线虫第 3期幼虫 ,在感染当天、感染后第 3d及第 7d分别腹腔注射大鼠抗 B7- 1和 /或 B7- 2单抗 ,以阻断共刺激信号。于感染当天 ,感染后第 7d和第 14 d,计数小鼠外周血液的嗜酸性粒细胞 ,于感染后第 14 d计数小肠绒毛嗜酸性粒细胞 ;EL ISA采用 Watanabe N方法测定 Ig E抗体。 结果 联合应用两种抗体抑制了外周血液及小肠组织的嗜酸性粒细胞增多 ;抑制了 Ig E抗体的产生。然而 ,单用抗 B7- 1或抗 B7- 2一种单克隆抗体 ,对嗜酸性粒细胞计数和 Ig E水平无明显影响。 结论 在巴西日圆线虫感染免疫中 ,T细胞的激活需要 B7- 1和 B7- 2共刺激分子的参与 ;而 B7- 1或 B7- 2单一分子即可提供足够的共刺激信号
【Abstract】 Objective To explore the effect of the constimulatory molecules B7 1 and B7 2 on the Th2 immune response to helminth infection. Methods BALB/c mice infected with Nipponstrongylus brasiliensis (Nb ) LⅢ were treated with anti B7 1 and /or anti B7 2 monoclonal antibodies on day 0, 3rd day and 7th day. Counting eosinophils of mice on day 0, 7th day and 14th day in peripheral blood and on 14th day in small intestine after infection. IgE was detected by Watanabe N ELISA. Results Combined treatment with anti B7 1 and anti B7 2 completely inhibited Nb induced eosinophil both in the blood and in the small intestine and suppressed IgE production. However, administration of either anti B7 1 or anti B7 2 alone had little effect on eosinophil and on the elevation of IgE levels. Conclusion B7 1 and B7 2 costimulation is required and either B7 1 or B7 2 can provide a sufficient costimulatory signal that lead to T cell activation and differentiation of function during a Th2 immune response to N. brasiliensis .
