中国南方汉族人群HLA-Cw座位基因多态性分析
Genetic polymorphism of HLA-Cw locus in a southern Chinese Han population analyzed by ARMS/PCR genotyping
【摘要】 目的 分析中国南方汉族人群HLA Cw座位基因多态性。方法 采用ARMS/PCR技术对 6 0名随机选择的南方汉族健康个体作HLA Cw基因分型。结果 共检出 12种HLA Cw等位基因或等位基因组 (allelegroup) ,其中HLA Cw 0 7、 0 1、 0 3为该人群最常见HLA Cw基因 ,频率之和为0 .6 932 ;证实该群体中HLA Cw 0 8频率为 0 .10 5 6 ,检出率明显高于血清学分型 ;检出HLA Cw 12 0 2、 12 0 3、 14、 15等 4种经典血清学分型不能鉴定的HLA Cw基因 ,频率之和为 0 .0 6 73。结论 提供了中国南方汉族人群HLA Cw座位基因频率正常值 ,证实ARMS/PCR作HLA Cw分型具有准确、快捷的优点
【Abstract】 Objective To explore the genetic polymorphism of HLA-Cw locus in Southern Chinese Han Population(SCHP). Methods HLA-Cw loci were genotyped for a panel of sixty randomly selected healthy SCHP individuals, using the ARMS/PCR technology described in 12th IHWC. Results Twelve HLA-Cw alleles or allele-groups were detected with HLA-Cw*07,*01,*03 as the most common HLA-Cw genes. HLA-Cw*08, previously thought to be rare in SCHP by serological typing, occupied a gene frequency of 0.1056. In addition, four kinds of non-serologically defined HLA-Cw* genes i.e. HLA-Cw*1202, *1203, *14, and *15 were found in the target population sharing 0.0673 of the total HLA-Cw frequency. Conclusion Normal HLA-Cw gene frequencies of SCHP were identified in the present study; The results also confirmed that ARMS/PCR was a reliable and fast method for HLA-Cw genotyping.
