急性白血病患者细胞周期蛋白E及依赖激酶抑制剂表达的意义
The significance of cyclin E expression and cyclin E-dependent kinase inhibitor in adult acute leukemia
【摘要】 目的 探讨细胞周期蛋白E(cyclinE)、细胞周期蛋白依赖激酶抑制剂 (p2 7KIP1)对成人初治急性白血病 (AL)发展及预后的作用。方法 用流式细胞术检测 6 0例AL及 17例对照cyclinE、p2 7KIP1、多药耐药基因蛋白p170表达及细胞周期分布 ;逆转录 聚合酶链反应检测 4 6例成人AL患者及 17例对照的骨髓cyclinE、p2 7KIP1、多药耐药蛋白的mRNA水平。结果 6 0例AL患者全周期及G2 /M期cyclinE表达均高于对照组 (P <0 0 1或 0 0 5 ) ;p2 7KIP1全周期表达亦均高于对照组 ,但G2 /M期差异无显著性 (P >0 0 5 ) ;cyclinE与p2 7KIP1的表达呈正相关 ;AL患者cyclinE高表达组缓解率(44 8% )低于正常表达组 (77 4 % ) (P <0 0 1) ;复发率 (92 3% )高于正常表达组 (41 7% ) (P =0 0 0 3) ;p2 7KIP1表达对AL缓解率、复发率的影响无统计学意义 (P >0 5 0 ,P =0 89)。结论 cyclinE在成人AL有异常高表达 ,并可能促使AL的发展 ,是AL缓解率、复发率的重要危险因素。
【Abstract】 Objective To evaluate the function and clinical vaule of cyclin E and p27 in adult acute leukemia(AL) Methods The protein expression of cyclin E, p27, p170 and cell cycle distribution was measured in the bone marrow samples of 60 adult patients with AL and 17 controls with flow cytometric analysis The mRNA levels of cyclin E, p27, MRP in 46 cases of AL and 17 controls were examined with reverse transcription polymerase chain reaction method Results The expression of cyclin E in AL patients was higher than that in controls ( P <0 01), so was the expression of p27 ( P <0 05) However in G 2/M phase the expression rate of cyclin E in AL patients was not so apparently different with that in control( P <0 05) so was that of p27( P >0 05) There was a positive correlation between the protein expression of cyclin E and p27 The remission rate (44 8%) in patients with over expression of cyclin E was lower than that (77 4%) in patients with normal expression of cyclin E ( P <0 01) The relapse rate (92 3%) in over expression group was higher than that (41 7%) in normal expression group ( P =0 003) No significant difference in remission rate or relapse rate was found between p27 over expression group and nomal expression group ( P >0 50, P =0 89) Conclusion Over expression of cyclin E may implicate occurrence and progression of AL Over expression of cyclin E is an important influential factor for remission rate and relpase rate in AL patients
