联苯双酯对家兔体内环孢菌素A的药动学影响

【摘要】 目的 :考察联苯双酯 (BFD)与环孢菌素A(CsA)在兔体内合用后联苯双酯对环孢菌素A动力学过程的影响。方法 :用高效液相色谱法测定 6只家兔单用环孢菌素A及合用联苯双酯后环孢菌素A的血药浓度并进行动力学参数的计算与比较。结果 :CsA与BFD合用后 ,CsA在家兔体内的血药浓度普遍降低 ,表观分布容积及清除率显著增大 (P <0 .0 5 ) ,AUC明显小于单用CsA(P <0 .0 5 ,P >0 .0 1)时的AUC ,其余的参数无统计学差异。结论 :BFD可明显降低CsA的血药浓度。

【Abstract】 OBJECTIVE To study changes in cyclosporine A(CsA) concentrations in whole blood and pharmacokinetics in rabbats before and after oral administration of bifendate(BFD).METHODS Six rabbits were first single administered with CsA,concentration of CsA in whole blood in different time were determined by high performance liquid chromatography(HPLC).after two days,BFD had been administered for four days,and the fourth day,CsA were oral administered,CsA were determined by the above method.RESULTS The diposition of CsA was conformed to two compartment model,after conbination with BFD,CsA concentrations in whole blood all decreased,AUC decreased( 0.05 > P > 0.01 ),Vc and Cl increased( 0.05 > P > 0.01 ).CONCLUSIONS BFD decreases CsA concentrations in whole blood.
