黑木耳酸性多糖(FⅡ)的分离、纯化及相对分子质量测定
Isolation purification and molecular weight determination o f acid polysaccharide of auricularia auricula-judae (FⅡ)
【摘要】 目的 :黑木耳酸性多糖 (FⅡ )的分离、纯化及分子质量测定。方法 :按文献方法 ,从黑木耳子实体提取得到酸性多糖(FⅡ ) ,采用 2 0 %H2 O2 溶液脱色 ,通过纸层析、柱层析、聚丙烯酰胺凝胶电泳法进行纯度鉴定 ,用小角激光光散射和膜渗透压法测定FⅡ的重均分子质量Mw 和数均分子质量Mn。结果 :FⅡ纯度较高 ,它的重均分子质量为 58.8× 1 0 4 ,数均分子质量为1 4 .3× 1 0 4 。结论 :多糖脱色最好用弱氧化剂 ,不要用强氧化剂 ,光散射法和膜渗透压法是测定多糖分子量的有效方法。
【Abstract】 OBJECTIVE To isolate acid polysaccharide auricularia auricula-judae (FⅡ) and determine i ts molecular weight.METHODS Acid polysaccharide(FⅡ) was isolated from auricularia auricula-judae,it was de colored with 20% H 2O 2,the purity was tested with paper chromatography,colum n chromatography and polyacrylamide gel electrophoresis.The weight average molec ular weight M w and number average molecular weight M n of FⅡwere measured b y low angle laser light scattering and membrane osmometry.RESULTS FⅡ is pure,its M w is 58.8 ×10 4 and its M n is 14.3 ×10 4.CONCLUSIONS Polysaccharide is decolored by weak oxidizers instead of strong ones,the method of laser light scattering and membrane osmometry is practical one for molecula r weight determination of polysaccharide.
