苯甲酰苯巴比妥对映体的毛细管电泳拆分研究
Study on chiral separation of benzonal by capillary electrophoresis
【摘要】 目的 首次建立抗癫痫新药苯甲酰苯巴比妥对映体的毛细管电泳拆分方法。方法 以 β环糊精 (β CD)为手性选择剂 ,考察了背景电解质 pH值、β CD浓度、样品介质、温度、操作电压和有机添加剂等因素对手性分离的影响 ,并提出了可能的手性识别机制。结果 在优化的电泳条件下 ,苯甲酰苯巴比妥对映体得到了基线分离 ,Rs 达 2 .0 4。结论 此法操作简单方便 ,手性分离效果好 ,可用于该新药的分离分析。
【Abstract】 OBJECTIVE A capillary electrophoresis method was developed for chiral separation of a new antiepileptic drug, benzonal, for the first time.METHODS β cyclodextrin (β CD) was used as a chiral selector. The effects of bugfer pH, concentration of β CD, sample medium,capillary temperature, applied voltage and organic additive on enantioseparation were examined respectively. The mechanism of chiral recognition by β CD in this system was discussed.RESULTS Under optimum conditions benzonal was completely separated with resolution as high as 2.04.CONCLUSION The proposed method was simple and rapid with high separation efficiency. It is recommended that this new method be used for chiral separation of benzonal.
