芝麻坏死花叶病毒(SNMV)部分cDNA合成及序列分析
Partial cDNA synthesis and sequence analysis of sesame necrotic mosaic virus
【摘要】 提取芝麻坏死花叶病毒RNA ,反转录合成cDNA ,对滞后重组质粒进行点杂交 ,得到阳性克隆并且测定序列。经计算机分析 ,获得 336 8个核苷酸 (nt)的序列。该序列含有一个完整的开放阅读框架 (ORF) ,该ORF由1134nt组成 ,推测编码 377个氨基酸 ,分子量为 4 0 .0 4 7× 10 3 道尔顿 (Da)。经比较它与Tombusviride科中Avreusvirus属石柑子潜隐病毒 (Pothoslatentvirus,PoLV)、Tombusvirus属病毒Cymbidiumringspotvirus(CRSV)外壳蛋白 (CP)氨基酸序列同源性分别为 4 0 .7%、4 0 .1% ,其起始氨基酸与Tombusvirus、Aureusvirus属另外的病毒CP起始氨基酸具有很高的保守性。根据这个完整ORF所编码蛋白的大小 ,与CRSV、PoLV病毒CP具一定的同源性 ,CP 5′端氨基酸的保守性 ,推测这个完整的ORF可能包含该病毒的外壳蛋白基因 ,并且这个病毒可能类似PoLV病毒 ,是Tombusviride科Aureusvirus属的一种新病毒
【Abstract】 The RNA of sesame necrotic mosaic virus (SNMV) was extracted from the purified virus particles and used as the template for random primer cDNA synthesis. The six positive clones which were detected by dot hybridization, were sequenced and identified the 3368 nucleotides by the computer analysis. The sequence included four encoding regions: one of which was consisted of 1134nt as a complete ORF and encodes 377 amino acids, the molecular mass was predicted to be 40.04kDa, the homology of its amino acid sequence with CP of CRSV and PoLV was 40.1% and 40.7% respectively. Its initial amino acids was consensus with the CP of viruses in Tombusvirus , Aureusvirus . According to these results, it is suggested that the complete ORF was SNMV CP gene and the virus was similar to PoLV be classfied into the family of Tombusviride , Aureusvirus .
