再灌注不同时间大鼠肝脏细胞凋亡的变化
The Change of Apoptosis during Normothermic Reperfusion on Rat Livers
【摘要】 目的 :通过大鼠肝脏热缺血再灌注模型 ,探讨大鼠肝脏细胞凋亡的变化。方法 :阻断大鼠肝脏门静脉左叶和中叶的分支 6 0min ,再灌注 0、2和 2 4h后 ,采用ELISA方法 ,测定肝组织中组蛋白相关的低分子量DNA片段 ,判断细胞凋亡的发生。结果 :发生缺血再灌注的肝叶中DNA的片段高于未发生损伤的肝叶 (P <0 .0 1) ;再灌注 2h后DNA的片段最多 (P <0 .0 1) ;同正常的肝脏相比 ,发生缺血再灌注损伤的肝脏中未被阻断血供的肝叶DNA的片段也较正常肝脏明显增加 (P <0 .0 1)。结论 :大鼠肝脏发生热缺血再灌注损伤时 ,细胞凋亡随之发生 ,且于再灌注早期最为明显
【Abstract】 Objective: By setting up the model of rat normonthermic ischemia and reperfusion(I/R) injury,we studied the change of apoptosis during this process Methods: We checked the liver tissue histo associated DNA segment which had been occluded blood supply for 60 min following by 0, 2 and 24 h reperfusion.The Apoptosis was determined by the liver tissue histo associated DNA segment using ELISA method. Results: Apoptosis in I/R injury lobe was more obvious than that in the lobe without I/R injury (P<0.01). Apoptosis after 2 h reperfusion was most obvious (P<0.01); the apoptosis in the lobe without I/R injury was more obvious than the normal liver tissues (P<0.01). Conclusion: Apoptosis was demonstrated during the occurrence of rat liver normothermic I/R injury,and was obvious at the early phase of reperfusion.
