【摘要】 目的 探讨LH基因免疫对前列腺增生模型鼠生殖内分泌的影响。方法RT-PCR法从雌性大鼠垂体细胞中RNA扩增大鼠的促黄体素基因(LH)cDNA片段约(461bp)并克隆至T载体,获得重组质粒T-LH,限制酶酶切,酶连接,构建PCDNA3.1(-)/HbsAg/LH,经脂质体转染纯化的重组质粒至COS细胞中。通过肌肉注射构建的 PCDNA3.1(-)/HbsAg/LH与前列腺增生大鼠,2个月内给药两次。结果 体外培养 COS细胞,发现HBsAg与LH连接物表达较好,肌肉注射前列腺增生模型鼠重组质粒后,模型鼠LH与T的含量明显低于对照组。结论PCDNA3.1(-)/HbsAg/LH可参与前列腺增生模型鼠生殖内分泌腺的调节。

【Abstract】 AIM The purpose is to investigate the effects of immunization against luteinizing hormone gene on endocrine of benign prostatic hyperplasic model rats. METHODS The rat luteinizing hormone gene(LH) was amplified from female rat pituitary RNA with RT-PCR and cloned into plasmid T vector to construct recombinant T-LH, which was then sequenced, the rat luteinizing hormone DNA fragment was cut from T-LH with EcoRI and Hindlll and inserted into eucaryotic expression vector PGDN A 3. 1(-)with HBSAg, transfected in COS cell by lipofectin reagant and in vitro and injected benign prostatic model rat. RESULTS The LH sequence of cloned rat luteinizing hormone was as same as that reported previously, and the eucaryotic plasmid expression recombinant of PCDN A3. 1(-)/HBsAg/LH showed a high level expression of recombinant protein, and has immune active, also after two mouths LH and T the benign prostatic hyperplasic model rats, which was injected the PGDN A3. 1(- )/ HBsAg/LH, was lower than the control group. CONCLUSION The PCDNA3. 1 (-)/HBsAg/LH may be the medicine for the beinign prostatic hyperplasic of rats.
